Case One: DNA 17mer Synthesis 
Purpose: 
Synthesize a strand of oligonucleotide DNA, Sequence: CTACGCCACGAGCTCCA, Synthesis amount 0.24mmol 
Experiments: 
1) Oligo Synthesizer: GoodScience RobotOligo 100 
2) Column: 6.3ml 
3) Carrier: HYS 350 
4) Deprotection reagent: coupling reagent, oxidation reagent, blocking reagent, anhydrous acetonitrile, ammonia 
5) Monomer:A、C、G、T 
6) 0.3bar Ammonia gas 
7) Mass Spectrometer: WATERS – THERMO

  RobotOligo Application 
Case Two: DNA 63mer Oligonucleotide fragment synthesis 
Purpose: 
Synthesize a single strand of ssDNA, 63mer (not listed as sequence is confidential), Synthesis amount 174μmol, Purity > 95% after purification. 
Experiments: 
1) Oligo Synthesizer: GoodScience RobotOligo 100 
2) Column: 6.3ml 
3) Carrier:Nitto H09002 
4) Reagents: deprotectant, activator, oxidant, thiolide, CAPA, CAPB, anhydrous acetonitrile, 20% DEA, ammonia. 
5) Monomer:A、C、G、T、U 
6) Protein Purification System: GoodScience Autopure 100 
7) Mass Spectrometer:ThermoFisher LTQ 
8) HPLC: GoodScience LC-3000 
Results Analysis: 
Purity of crude product is 46.3%, and purity reached 97.8% after one-step reverse-phase chromatography purification. Finally, the molecular weight of 
the target product is 19344, which is matching with the theoretical molecular weight of the target product (19342.64). 

  RobotOligo Application 
Case Three: RNA 21mer Synthesis 
Purpose: 
Synthesize a single strand of RNA, 21mer (not listed as sequence is confidential), Synthesis amount 245μmol. 
Experiments: 
Oligo Synthesizer: GoodScience RobotOligo 100 
Results Analysis: 
Purity of crude product is 84.84%, and purity reached 94.37% after purification. Molecular weight measured by mass spectrometry is 7097.7, which is 
matching with theoretical value (7096.8).

Case Four: RNA 76mer Synthesis 
 Purpose: 
Synthesize a single strand of RNA, 76mer (not listed as sequence is confidential), Synthesis amount 70μmol. 
 Experiments: 
Oligo Synthesizer:GoodScience RobotOligo 100 
 Results Analysis: 
Purity of crude product is 69.61%, and purity reached 93.93% after purification. Molecular weight measured by mass spectrometry is 25133, which is 
matching with theoretical value (25131.2).
